57 Abstract AB [DE] Isolierte, gereinigte Vorläuferzellen mit neuronalen oder glialen Eigenschaften aus embryonalen Stammzellen, enthaltend höchstens etwa 15% primitive embryonale und nicht-neutrale Zellen, erhältlich durch folgende Schritte: a) kultivieren von ES-Zellen zu Embryoid Bodies, b) kultivieren der Embryoid Bodies zu neutralen Vorläuferzellen, c) proliferieren der neuralen Vorläuferzellen in einem Wachstumsfaktor-haltigen serumfreien Medium, d) proliferieren der neutralen Vorläuferzellen aus Schritt c) in einem weiteren Wachstumsfaktor-haltigen serumfreien Medium und isolieren der gereinigten neutralen Vorläuferzellen und e) proliferieren der neutralen Vorläuferzellen aus Schritt d) in einem weiteren Wachstumsfaktor-haltigen serumfreien Medium und isolieren der gereinigten Vorläuferzellen, mit neuronalen oder glialen Eigenschaften, oder a') kultivieren von ES-Zellen zu Embryoid Bodies, b') kultivieren der Embryoid Bodies zu neutralen Vorläuferzellen, c') proliferieren der neutralen Vorläuferzellen in einem Wachstumsfaktor-haltigen serumfreien Medium, d') proliferieren der neutralen Vorläuferzellen aus Schritt c') in einem weiteren Wachstumsfaktor-haltigen serumfreien Medium zu Sphäroiden mit neuronalem und glialem Differenzierungspotential und isolieren der neuralen Sphäroide und e') proliferieren der neutralen Sphäre aus Schritt d') in einem Wachstumsfaktor-haltigen serumfreien Medium bis zur Ausbildung eines aus glialen Vorläuferzellen bestehenden Zellrasens und isolieren der gereinigten Vorläuferzellen mit glialen Eigenschaften.
[EN] A process for producing neuronal or glial progenitor cells, comprising: (a) culturing embryonic stem cells to form embryoid bodies; (b) culturing the embryoid bodies to form neural progenitor cells; (c) proliferating the neural progenitor cells in a serum-free medium containing growth factor(s); (d) further proliferating the neural progenitor cells in a serum-free medium containing growth factor(s) and isolating the neural progenitor cells; and (e) further proliferating the neural progenitor cells in a serum-free medium containing growth factor(s) and isolating the neural progenitor cells. Independent claims are included for the following: (1) a process for producing glial progenitor cells, comprising: (a) culturing embryonic stem cells to form embryoid bodies; (b) culturing the embryoid bodies to form neural progenitor cells; (c) proliferating the neural progenitor cells in a serum-free medium containing growth factor(s); (d) further proliferating the neural progenitor cells in a serum-free medium containing growth factor(s) form spheroids with neuronal and glial differentiation potential; and (e) proliferating the spheroids in a serum-free medium containing growth factor(s) until a lawn of glial progenitor cells is formed, and isolating the cells; (2) neuronal or glial progenitor cells that are produced as above and include no more than 15% of primitive embryonic cells and non-neural cells; and (3) a cell library comprising autologous and nonautologous cells as above.
